Analysis (determination) of products of metabolism of microorganisms

Analysis of microbial metabolic products using HPLC and gas chromatography. Data on metabolic products (organic acids, mono- and polyatomic alcohols, esters and gaseous products) are among the important taxonomic characteristics of a number of groups of bacteria and archaea and are necessary for the description of new species and genera. The determination of the above properties also makes it possible to evaluate the possibility of using the studied microorganisms as producers of the corresponding compounds.

Determination of chemotaxonomic characteristics of microbial strains

Determination of taxonomic characteristics of strains of microorganisms (bacteria, archaea, fungi, yeast) required to describe new species and genera of microorganisms in accordance with international standards. The list of such characteristics includes: the isomer of diaminopimelic acid of the cell wall, the composition of amino acids of the cell wall, the composition of sugars of whole cells/cell walls, the type of mycolic acids, the composition of fatty acids, the composition of isoprenoid quinones of the respiratory chain (menaquinones, ubiquinones), the composition of phospholipids, the nucleotide composition of DNA.

Identification of microbial strains

Identification of microbial strains (the MALDI method of mass spectrometry) using the manufacturer's database (Brooker) and local databases created at the CCP VKM. The method is based on comparing the mass spectra of a mixture of components of destroyed cells (the components of the MALDI spectra in the mass range 2000-20000 m/z correspond to polar cytoplasmic proteins, mainly ribosomal). The advantages of the method are the speed and small amount of material for analysis (one colony is enough).
