The OBK of the IGF RAS provides comprehensive scientific research in the field of biotechnology of higher plants. The OBK of the IGF RAS includes three sections: preparatory, production and finished product receiving section, equipped with both domestic and imported equipment.

At the preparatory site, work is carried out on air purification; preparation and sterilization of dishes and equipment; preparation and sterilization of cultivation media; maintenance in collections of producing crops; preparation of selected strains for hardware cultivation. The site is equipped with a set of distillers and steam autoclaves, washing equipment, sterilization and drying plants, climate chambers with controlled environmental conditions and stationary circular rockers for long-term cultivation of producer strains in flasks with a speed control system (for flasks 0.25 — 2.00 liters), laminar flow cabinets, as well as analytical equipment. The production site is equipped with fermentation plants with a volume from 2 to 2500 liters, in which the actual hardware cultivation of producer crops is carried out in laboratory, pilot and semi-industrial plants while simultaneously monitoring the processes of growth and biosynthesis. At the site of receiving finished products, plant cells and adventitious roots are separated from the culture liquid, dried, dry biomass is analyzed, packaged and stored. The sector is equipped with a set of filtration and drying equipment that allows working with various volumes of cell suspensions and adventitious roots, as well as appropriate analytical equipment for chemical analysis of the products (HPLC, HPLC-MS, spectrophotometers).

According to the set of parameters, the IGF RAS is a unique installation for the development of biotechnologies for the industrial production of biological products with specified properties for the needs of the food, pharmaceutical, and cosmetic industries based on cell cultures and organs of higher plants, regardless of environmental conditions. The characteristics of the available technological equipment make it possible to create an environmentally safe and economically profitable production scheme when scaling laboratory developments.

  1. Methods for evaluating the growth of cell cultures and organs of higher plants (determination of growth parameters, viability, duration of phases of the growth cycle, construction of growth curves)
  2. Methods of qualitative and quantitative assessment of the content of secondary metabolites in cultures of cells and organs of higher plants (HPLC, NMR, HPLC-MS, spectrophotometry)
  3. Methods for obtaining adventitious roots, callus and suspension cell cultures of higher plants. Methods for assessing the respiratory metabolism of suspension cultures of higher plant cells
  4. Methods for evaluating the biological activity of extracts from the biomass of cells and adventitious roots of higher plants

Research directions

Creation and development of industrial biotechnologies for obtaining valuable BAS based on cell cultures and organs of higher plants.

+
A scaling scheme for growing suspension cell cultures (in particular Taxus wallichiana, Dioscorea deltoidea, Panax japonicus and Polyscias filicifolia) in the weaning-topping mode in bubbling bioreactors in the range of 20 l—75 l—630 l has been developed and optimized. Pilot industrial regulations have been developed. The data obtained make it possible to characterize the biomass of plant cell cultures as an environmentally friendly, safe plant raw material and recommend it for use in functional food products, cosmetics, pharmaceuticals and feed.

Obtaining and comprehensive study of the characteristics of crops of adventitious roots of higher plants

+
As part of the work on the Megagrant of the Government of the Russian Federation No. 075-15-2019-1882, work has begun and is continuing to obtain and conduct a comprehensive study of the characteristics of adventitious root cultures using protocols and consultations of the leading scientist Kee-Yoeup Paek (South Korea). At the moment, the staff of the UNU OBK IGF RAS has obtained and maintained 11 lines of adventitious roots of 11 species in parallel on solid and liquid media (see the list of objects).
